Use of Targeted Nanosomes to Detect Damaged Cartilage in Collagen Induced Arthritis

چکیده
INTRODUCTION: Most of the therapeutic compounds used in the treatment of rheumatoid arthritis (RA) and osteoarthritis (OA) are given systemically. In an effort to more specifically target diseased and damaged areas of cartilage and to significantly reduce the risk of systemic side effects, we have devised a theranostic method of drug delivery using immunoliposomes. Unilamellar vesicles (ULV) in the size range of 200400 nm and labeled with a near-infrared emitting dye (NIF) were conjugated to the monoclonal antibody to type II collagen (MabCII) and injected into mice with collagen induced arthritis (CIA), a model of inflammatory arthritis, to determine if the bound nanosomes would be easily detected using in vivo imaging techniques.

Noninvasive visualization of early osteoarthritic cartilage using targeted nanosomes in a destabilization of the medial meniscus mouse model
Background Early stage osteoarthritis (OA) is clinically asymptomatic due to the avascular and the aneural nature of the cartilage tissue. Nevertheless, early detection of cartilage tissue is critical in order to impede the progression of OA. Hence, in order to develop effective preventive therapy for OA, diagnosis in the early stages is necessary. Methods To achieve this goal, we have develo...
متن کاملH3K27me3 demethylases regulate in vitro chondrogenesis and chondrocyte activity in osteoarthritis
BACKGROUND Epigenetic changes (i.e., chromatin modifications) occur during chondrogenesis and in osteoarthritis (OA). We investigated the effect of H3K27me3 demethylase inhibition on chondrogenesis and assessed its utility in cartilage tissue engineering and in understanding cartilage destruction in OA. METHODS We used a high-content screen to assess the effect of epigenetic modifying compoun...
متن کاملT Cell Vaccination as a Tool in the Treatment of Collagen Induced Arthritis in Albino Rats
Background: The effectiveness of T cell vaccination has been demonstrated in a variety of animal models of both induced and spontaneous autoimmune diseases. Objective: The purpose of this study was to test the T cell vaccination protocol to treat and prevent collagen induced arthritis (CIA) in a rheumatoid arthritis model. Methods: CIA was induced by an intradermal injection of an artheritogen ...
متن کاملIL-4 Can Inhibit IL-17 Production in Collagen Induced Arthritis
Background: IL-4 is a cytokine that induces differentiation of naive helper T cells into Th2 cells. Once activated by IL-4, Th2 cells subsequently produce additional IL-4. Objective: To examine the effect of IL-4 on IL-17 production and its effect in Collagen- Induced Arthritis (CIA) mice. Method: In this study, a chicken collagen-II-induced experimental arthritis (CIA) model was used in DBA/1 ...
